  • Ⅱ型糖尿病兔大脑皮质和海马神经元的神经丝蛋白表达

    Objective:To investigate the morphological changes of the neuronal neurites in diabetic rabbit brain. Methods: Twenty- four New Zealand White rabbits were divided into 2 groups: control group and type Ⅱ diabetic group induced by high - carbohydrate and high- fat diet. The levels of blood sugar and insulin were detected at week 0(w0), w4, w8, w13, w18, w23 and w28. Brain tissue was stained by Nissl staining and immunolistochemistry with a specific antibody to neurofilament proteins. Result: In diabetic rabbits, the amount of large pyramidal neuron was significantly reduced, and neuronal neurites became swollen, whorled, disrupted and changed in caliber. In hippocampus CA1 region neurofilament staining was very weak. Conclusion: Neurotoxicity of chronic hyperglycemia might be relevant to vascular chronic complications, which affected the expression of NF and led to neurophysiological and structural changes in the brain of rabbits with type Ⅱ diabetes.

    Objective To build an amimal model of unstable atherosclerotic plaques.Methods Sixty-four New Zealand White rabbits were randomly divided into two groups: group A(n= 54)and B(n= 10). Rabbits in group A underwent balloon-induced arterial wall injury and then were given a diet of 1% cholesterol; rabbits in group B were only given 1% cholesterol.

    Objective To identify the characteristic features of unstable atherosclerotic plaques with intravascular ultrasound(IVUS).Methods Seventeen New Zealand White rabbits underwent balloon-induced abdominal aortic wall injury and were given a diet of1% cholesterol for 8 weeks. The aortic segments rich in plaques were then incubated transluminally with recombinant adenovirus carrying a p53 transgene.

    BACKGROUND: Studies have shown that sodium hyaluronate inhibits cartilage damage in osteoarthritis and accelerates regeneration of cartilage cels, to stabilize and repair the articular cartilage.
    OBJECTIVE:To investigate the therapeutic effect of sodium-rich plasma combined with platelet-rich plasma (PRP) on rabbit knee osteoarthritis.
    METHODS:Forty New Zealand white rabbits were randomly divided into five groups, control group, combined group, sodium hyaluronate group, PRP group and model group, and then an osteoarthritis model of the right knee was made in each rabbit. After modeling, sodium hyaluronate+PRP, sodium hyaluronate, autologous PRP and normal saline were givenviathe knee joint cavity in the latter four groups, respectively, once a week for 5 weeks. The control group received no treatment, as normal controls. At 1 week after treatment, ELISA assay was used to detect serum interleukin-1, interleukin-6, tumor necrosis factor-α levels, and changes of the articular cartilage were observed under a light microscope.
    RESULTS AND CONCLUSION:Compared with the control group, the levels of interleukin-1, interleukin-6 and tumor necrosis factor-α were al increased in the other four groups (P < 0.01). Compared with the model group, the levels of interleukin-1, interleukin-6 and tumor necrosis factor-α were lowered significantly in the combined, sodium hyaluronate and PRP groups (P< 0.01 orP< 0.05), and the most significant decline was in the combined group. Articular cartilage damage was severest in the model group and mildest in the combined group. Experimental findings indicate that intra-articular injection of sodium hyaluronate+PRP can reduce inflammation and protect the articular cartilage in knee osteoarthritis, which is better than a single drug injection.

    To observe the effects of diabetogenic (high fat high sucrose, lacking choleserol) diet on atherogenesis in New Zealand white rabbits. Two groups of New Zealand white rabbits received regular rabbit chow (the normal control), or high fat high sucrose diet for 4 months. The levels of plasma total cholesterol, HDL cholesterol, triglycerides, insulin, and glucose were investigated, the areas of fatty streak of the aortae were measured after staining with Sodan IV, and the aortic, coronary specimens were observed with light and electron microscopies. The plasma glucose, triglycerides, and total cholesterol were increased significantly by high fat high sucrose feeding. At the end of 4 months, the early charateristics of atherosclerosis were present in the animals' vascular specimens. Our findings suggest that high fat high sucrose feeding can induce hyperglycemia, hypertriglyceridemia and atherosclerosis in New Zealand white rabbits, and this could be a potential animal model for studying the mechanisms of diabetes-accelerated atherosclerosis. This study raised a question: What is the mechanism by which high fat high sucrose feeding induces atherosclerosis?. The related hypothesis was given in this article.

    Abstract] AIM and METHODS:The recent work from our laboratory showed that ventral septal area (VSA) played a negative-regulatory central role in thermoregulation during endogenous pyrogen-induced fever. In order to further investigate the role of VSA in the antipyretic mechanism, we observed the effects of electrical stimulation of VSA on firing characteristics of thermosensitive neurons in preoptic anterior hypothalamus (POAH) by using extracellular microelectrode technique on 32 New Zealand white rabbits treated with interleukin-1 β (IL-1β) intracerebroventriculary (ICV). RESULTS:(1)Injection of IL-1β decreased discharging rate of warm-sensitive neurons in POAH. Electrical stimulation of VSA remarkably decreased thermosensitive coefficient of warm-sensitive neurons. (2)Injection of IL-1β caused increase in discharging rate of cold-sensitive neurons in POAH. Electrical stimulation of VSA remarkably increased thermosensitive coefficient of cold-sensitive neurons. CONCLUSTION:VSA may have an antipyretic effect through affecting the firing characteristics of thermosensitive neurons in POAH during IL-1β-induced fever.
