    Previous studies have shown that vagus nerve stimulation can improve the prognosis of trau-matic brain injury. The aim of this study was to elucidate the mechanism of the neuroprotective effects of vagus nerve stimulation in rabbits with brain explosive injury. Rabbits with brain ex-plosive injury received continuous stimulation (10 V, 5 Hz, 5 ms, 20 minutes) of the right cervical vagus nerve. Tumor necrosis factor-α, interleukin-1βand interleukin-10 concentrations were detected in serum and brain tissues, and water content in brain tissues was measured. Results showed that vagus nerve stimulation could reduce the degree of brain edema, decrease tumor necrosis factor-αand interleukin-1βconcentrations, and increase interleukin-10 concentration after brain explosive injury in rabbits. These data suggest that vagus nerve stimulation may exert neuroprotective effects against explosive injury via regulating the expression of tumor necrosis factor-α, interleukin-1βand interleukin-10 in the serum and brain tissue.

    Approximately 50-70% of patients experience incision-induced mechanical nociception after sur-gery. However, the mechanism underlying incision-induced mechanical nociception is stil unclear. Interleukin-10 and brain-derived neurotrophic factor are important pain mediators, but whether in-terleukin-10 and brain-derived neurotrophic factor are involved in incision-induced mechanical no-ciception remains uncertain. In this study, forty rats were divided randomly into the incision surgery (n=32) and sham surgery (n=8) groups. Plantar incision on the central part of left hind paw was performed under anesthesia in rats from the surgery group. Rats in the sham surgery group re-ceived anesthesia, but not an incision. Von Frey test results showed that, compared with the sham surgery group, incision surgery decreased the withdrawal threshold of rats at 0.5, 3, 6 and 24 hours after incision. Immunofluorescence staining in the dorsal root ganglia of the spinal cord (L 3-5 ) showed that interleukin-10 and brain-derived neurotrophic factor were expressed mainly on smal-and medium-sized neurons (diameter<20μm and 20-40μm) and satel ite cells in the dorsal root ganglia of the spinal cord (L 3-5 ) in the sham surgery group. By contrast, in the surgery group, high expression of interleukin-10 and brain-derived neurotrophic factor appeared in large-sized neurons (diameter>40μm) at 6 and 24 hours after incision surgery, which corresponded to the decreased mechanical withdrawal threshold of rats in the surgery group. These experimental findings suggest that expression pattern shift of interleukin-10 and brain-derived neurotrophic factor induced by inci-sion surgery in dorsal root ganglia of rats was closely involved in lowering the threshold to me-chanical stimulus in the hind paw fol owing incision surgery. Pain-related mediators induced by in-cision surgery in dorsal root ganglia of rats possibly underlie mechanical nociception in ipsilateral hind paws.

    Aim:To investigate if interleukin-6 (IL-6), interleukin-8 (IL-8), interleukin-10 (IL-10), interferon-gamma (IFN-γ) or tumor necrosis factor-alpha (TNF-α) are able to stimulate the level of lipid peroxidation of sperm membranes, either alone or in the presence of leukocytes. Methods: Semen samples from normozoospermic donors were prepared by density gradient. The sperms were exposed to the indicated cytokines, at physiological and infection-inflammation concentrations, in the absence or presence of leukocytes. Lipid peroxidation of the sperm membranes was determined by measuring malondialdehyde (MDA) and 4-hydroxialkenals (HAE) formation. Results: TNF-α, IL-8 and IFN-γ increased the level of sperm membrane lipid peroxidation when tested at physiological concentrations. At infectioninflammation concentrations, only IL-8 was able to produce a higher effect. When assayed in the presence of leucocytes,IL-8 and TNF-α showed a higher effect at infection-inflammation concentrations than at physiological concentrations.Finally, IL-8 showed a higher effect in the presence of leukocytes than in their absence at both physiological and infection-inflammation concentrations. TNF-α also showed a higher effect when assayed in the presence of leukocytes than in their absence, but only at infection-inflammation concentrations. There was no effect of IL-6 or IL- 10 in any of the tested conditions. Conclusion: Several pro-inflammatory cytokines at physiological concentrations increase the level of lipid peroxidation of sperm membranes, which could be important for the sperm fecundation process. However, infection-inflammation concentrations of some cytokines, such as IL-8 and TNF-α, either alone or in the presence of leukocytes, could drive the lipid peroxidation of the spermatozoa plasma membrane to levels that can affect the sperm fertility capacity.

  • 他米巴罗汀对牙龈卟啉单胞菌抗原刺激RAW264.7细胞的影响

    目的 探讨他米巴罗汀(Tamibarotene,Am80)对经牙龈卟啉单胞菌(Porphyromonas gingivalis,P.gingivalis)抗原刺激后的小鼠单核巨噬细胞RAW264.7的影响.方法 采用P.gingivalis抗原刺激RAW264.7细胞,分别用不同浓度的Am80进行干预;相差显微镜下观察各组细胞生长情况;MTS法检测不同浓度Am80对抗原刺激RAW264.7细胞的增殖情况;酶联免疫吸附法(ELISA)检测细胞培养上清中抗炎因子IL-10的含量.结果 10 nmol/L Am80能够显著抑制P.gingivalis抗原刺激RAW264.7细胞的增殖分化(P<0.001);Am80(10 nmol/L)致抗原刺激的细胞IL-10的分泌量显著增加(P<0.05);倒置相差显微镜下观察显示Am80抑制抗原刺激细胞的增殖分化.结论 Am80(10 nmol/L)时可显著促进P.gingivalis抗原诱导小鼠巨噬细胞分泌抗炎因子IL-10,并可能由此抑制P.gingivalis抗原诱导的炎症反应.

  • 肺癌患者肺泡巨噬细胞和外周血单核细胞表达IL-10及其临床意义

    目的探索肺癌患者肺泡巨噬细胞(AM)和外周血单核细胞(PBMC)分泌白介素-10(Interleukin-10, IL-10)的能力,了解肺癌患者局部及全身的细胞免疫功能状态.方法从57例肺癌患者、33例良性肺病患者、12例正常人的支气管肺泡灌洗液及外周血中分别获取肺泡巨噬细胞(AM)及外周血单核细胞(PBMC),用ELISA法测定其培养上清液中IL-10含量.结果 (1)肺癌组AM及PBMC分泌IL-10的水平显著高于正常人组和良性肺病组(P<0.001).(2)Ⅳ期肺癌患者PBMC分泌IL-10水平显著高于Ⅰ+Ⅱ期和Ⅲ期患者(P<0.001),Ⅲ、Ⅳ期患者AM分泌IL-10水平显著高于Ⅰ+Ⅱ期(P<0.001);小细胞未分化癌患者PBMC、AM分泌的IL-10水平显著高于非小细胞肺癌患者(P<0.01);肿瘤直径越大、KPS评分越低,PBMC、AM分泌IL-10水平则越高;生存期≥2年的肺癌患者PBMC、AM分泌IL-10的水平显著低于生存期<2年者(P<0.01).结论肺癌患者全身及局部都存在较高水平的IL-10,IL-10在肺癌的恶化进程方面可能发挥着重要作用.检测IL-10水平可作为评价肺癌患者细胞免疫状态、预后判断的有效途径.
