Abstract:

目的: 研究细胞在体内的生物分布、持久性和存活率是开发细胞治疗癌症的瓶颈。使用全氟化碳 (PFC) 纳米乳剂进行离体细胞标记，与 19 F MRI 检测配对，是体内细胞产物检测的非侵入性方法。淋巴细胞小，弱吞噬限制性 PFC 标记水平和 MRI 敏感性。为了增强标记，我们设计了显示细胞穿透肽的 PFC 纳米乳成像探针，即人类免疫病毒的反式激活转录序列 (TAT)。我们报道了制备 TAT 助表面活性剂的优化合成方案，以补充 PFC 纳米乳制剂中常用的表面活性剂。方法: 我们用纳米乳对原代人嵌合抗原受体 (CAR) T 细胞进行离体标记。使用电子显微镜和共聚焦成像验证细胞内标记。为了检测体内信号增强，将标记的 CAR T 细胞瘤内注射到荷侧腹胶质瘤的小鼠体内。结果: 通过将 TAT 掺入纳米乳中, 每个 CAR T 细胞达到 ~ 1012 个氟原子的标记效率，与不含 TAT 的纳米乳液相比，增加了> 8 倍，同时保持了高细胞活力 (~ 84%)。流式细胞术表型分析显示，用 TAT 纳米乳标记后 CAR T 细胞不变，体外肿瘤细胞杀伤实验显示细胞毒功能完整。从 TAT 标记的 CAR T 细胞中检测到的 19 F MRI 信号比没有 TAT 的 PFC 标记的细胞高 8 倍。结论: 提出的肽-PFC 纳米乳合成方案可显著增强细胞标记和成像灵敏度，可推广用于其他靶向成像探针。
