Abstract:

猪生长激素 (pGH) 是由垂体分泌的一类肽类激素，能显著提高猪的生长和饲料利用率。然而，它在体外是不稳定的和挥发性的。饲养家畜时需要将其包裹在脂质体中，其高昂的成本极大地限制了其在养猪业中的应用。因此，我们试图在毕赤酵母中表达细胞内可溶性蛋白 pGH，并以猪部分破壁的酵母菌为原料, 可以在肠道内直接释放 pGH，以防在肠道内降解，成本低。为了提高 pGH 蛋白在毕赤酵母中的胞内可溶性表达和体外稳定性，我们对 pGH 基因进行了优化，并从 E. 大肠杆菌和毕赤酵母分别与 pGH 共表达。此外，我们还探索了机械破碎和发酵的条件。结果表明，经基因优化后与毕赤酵母 Ssa1-Sis1 分子伴侣共表达，细胞内可溶性 pGH 蛋白表达量显著增加。同时，确定了毕赤酵母部分破壁发酵的最佳条件, 结果表明，在部分破壁发酵条件下，与 Ssa1-Sis1 共表达的优化 pGH 蛋白胞内表达量可达 340 mg/L。动物实验验证了优化后的 pGH 蛋白与 Ssa1-Sis1 共表达对仔猪生长的促进作用最好。我们的研究表明，Ssa1-Sis1 可以增强毕赤酵母中 pGH 蛋白的胞内可溶性表达，并且酵母的部分破壁可以阻止 pGH 在体外的降解。在肠内有针对性地释放，有效地发挥其生物学功能。本研究为有效降低成本提供了新思路，为体外不稳定物质的临床应用奠定了理论基础。
