AP4B1 的新变异引起 1 例中国患者痉挛性四肢瘫、中度精神运动发育迟缓和热性惊厥: 病例报告。

Abstract:

引言: AP4B1 基因编码接头蛋白复合物-4 (AP4) 的一个亚基，AP4 是细胞内蛋白质运输的一个组成部分，在神经元中起重要作用。AP4B1 的双等位基因突变引起常染色体隐性遗传性痉挛性截瘫-47 (SPG47)。病例介绍: 在这里我们介绍了 1 例痉挛性四肢瘫、中度精神运动发育迟缓和热性惊厥加的中国患者。脑 mri 表现为幕上脑室扩张，胼胝体后、压部薄。通过药物治疗和康复方案，患者进展甚微。全外显子组测序鉴定出 AP4B1 基因新的复合杂合截断变异 c.1207C> t (p.Gln403 *) 和 c.52 _ 53 delAC (p.Cys18Glnfs * 7)。AP4B1 的因果突变迄今已在 22 个家系的 29 个个体中报道，其中大部分是纯合突变。结论: 我们的研究丰富了 spg47 的遗传和表型谱。早期发现、诊断和适当治疗通常会增加患者生活质量改善的机会。
